Separation-based TRF assays

Separation based TRF assays, also known as Dissociation-Enhanced Lanthanide Fluorescent Immunoassays, offer a superior alternative to traditional ELISA technology. These assays are significantly more sensitive, have a broader dynamic range and are more robust than enzyme-based assays. Separation assays require a coated plate as the solid phase. The plates can be coated with cells, antibodies, streptavidin or other suitable tags. The principle of Dissociation-Enhanced. Lanthanide Fluorescent Immunoassays are shown below:

Separation based TRF

  • The ligand is labelled with a non-fluorescent Eu chelate.
  • Following binding of ligand to a receptor immobilised on solid phase unbound labelled reagent is washed away.
  • Enhancement buffer is added causing release of Eu(III) from nonfluorescent chelate to form a new, highly fluorescent chelate.
  • Measurement.

TRF assays

TRF requires a suitable acceptor molecule for the Europium chelate. FRET uses two fluorophores, a donor and an acceptor. Excitation of the donor by a suitable energy source triggers an energy transfer to the acceptor if they are within a given proximity to each other. The acceptor in turn emits light at its given wavelength.
